Tolazoline is structurally and pharmacologically related to phentolamine (see also Phentolamine). Experience with tolazoline in pregnancy is limited. The Collaborative Perinatal Project monitored two 1st-trimester exposures to tolazoline plus 13 other patients exposed to other vasodilators (1). From this small group of 15 patients, 4 malformed children were produced, a statistically significant incidence (p<0.02). It was not stated whether tolazoline was taken by any of the mothers of the affected infants. Although the data serve as a warning, the number of patients is so small that conclusions as to the relative safety of this drug in pregnancy cannot be made.
