Name: LEUPROLIDE
Class: Antineoplastic/Hormone
Risk Factor: XM
Fetal Risk Summary
Leuprolide is a synthetic nonapeptide analogue of naturally occurring gonadotropin-releasing hormone that inhibits the secretion of gonadotropin when given continuously and in therapeutic doses.
Leuprolide causes a dose-related increase in the incidence of major malformations in pregnant rabbits, but not in rats (1). The most frequently observed malformations in rabbits were vertebral anomalies and hydrocephalus (J.D. Miller, personal communication, Tap Pharmaceuticals, Inc., 1992). The doses tested were 1/300–1/3 of the typical human dose. Increased fetal mortality and decreased fetal weights were observed in both animal species with the higher test doses.
In humans, spontaneous abortions or intrauterine growth retardation are theoretically possible because leuprolide suppresses endometrial proliferation. The risk of these adverse outcomes is considered greater than the risk of congenital malformations because the affected organs in animal studies do not depend on the presence of gonadal steroids for normal development (J.D. Miller, personal communication, Tap Pharmaceuticals, Inc., 1992).
The manufacturer is maintaining a registry of inadvertent human exposures during pregnancy to leuprolide and currently has more than 100 such cases (J.D. Miller, personal communication, Tap Pharmaceuticals, Inc., 1992). No cases of congenital defects attributable to the drug have been reported, although the numbers are too small to draw conclusions as to the risk for perinatal mortality, low birth weight, or teratogenicity.

Q&A about Leuprolide
I had emergency laparotomy/laparscopy 9 days ago to discover I had a severe case of endo. My physician is highly recommending Lupron as treatment for 6 months. It is to trick my body in thinking I'm menopausal. If it's successful, It can help my recovery and reduce pain by stopping my periods basically that I know. When I see my doc in a few days to start the treatment, I'm not sure how long after the first treatment series if there'll be a continuance. I'm 19 years old and overall healthy. I wanted a family in 10 years, but now I'm lucky to have 5. Will the treatment affect future HEALTHY pregnancies? How long will I have to be off the treatment b4 I am able to even get pregnant much less a healthy one? Also I only have my right tube left (which was unblocked during surgery b/c of endo took great damage) My left tube ruptured with an ectopic 3 months ago. So my chances now are becoming lower and lower it seems though I've been maintaining hope.Any advice? Info pertaining NEthing
Lupron will help if he recommend L it is because you have fibroid inside your tubes and since you wanted chidren it is better to give you shot of lup than removed all of them (you might lost your tube)you can try to get Preg after 45 days of your last shot
