Calcitriol is one of three physiologically active forms of vitamin D (see Vitamin D). Calcitriol is teratogenic in rabbits dosed with 0.08 and 0.3 mg/kg (about 2 and 6 times the maximum recommended human dose based on mg/m2 [MRHD]) (1). Defects observed included external and skeletal malformations. The higher dose (0.3 mg/kg/day) given on gestation days 7 to 18 also increased maternal mortality, decreased mean fetal body weight, and reduced the number of newborn surviving to 24 hours. No teratogenic effects were observed in rats treated with doses up to 0.45 mg/kg (about 5 times the MRHD) (1). However, hypercalcemia was measured in the offspring of rats given 0.08 and 0.3 g/kg/day (about 1 and 3 times the MRHD) (1).
The manufacturer cites a case of mild hypercalcemia during the first 2 days of life in an infant who was exposed to a maternal dose of 17 to 36 mg/day (approximately 17 to 36 times the maximum recommended dose) during pregnancy (1). The hypercalcemia resolved by the third day.

Both are effective in treating hypoparathyroidism and chronic renal failure. Calcitrol also treats rickets.

Carry out the method for liquid chromatography, in subdued light using the following solutions.
For capsules containing 0.25 µg or less, use the mixed contents of 10 capsules as solution (1). For capsules containing more than 0.25 µg, prepare solution (1) by diluting a quantity of the mixed contents of 10 capsules containing 1.5 µg of Calcitriol to 1 ml with the mobile phase.
Solution (2) contains 0.00015% w/v of calcitriol EPCRS in the mobile phase.
The chromatographic procedure may be carried out using
(a) a stainless steel column (25 cm × 4.6 mm) packed with stationary phase A (5 µm) (Lichrosorb Si60 is suitable),
(b) a mixture of 1 volume of propan-1-ol , 2 volumes of methanol , 40 volumes of hexane and 60 volumes of ethyl acetate as the mobile phase with a flow rate of 1.2 ml per minute and
(c) a detection wavelength of 265 nm.
If necessary, adjust the composition of the mobile phase so that the principal peak in the chromatogram obtained with solution (1) is clearly separated from the tail of the peak due to the excipient.

Doctors & informed people plse, help, black stool!?, My aged father, suffering from old age and Alzheimer's disease, is sometimes incapable of controlling his defecation, and is often excreting black stool. He is under treatment with the following medicine: Aricept, Citalopram, Propronolol, Aspirin (100mg), Calcitriol, multivitam+mineral, royal jelly, etc. Is the stool colour a sign of bleeding or something else? What do you recommend?
First I recommend that a small sample of his stool, collected in a dry container be submitted to the doctor for quick testing for blood a stool Guaiac test. Black stool, if positive for blood, usually means upper digestive system bleeding, like blood from the stomach or small intestine, not from the lower bowel or rectum where the blood is usually a brighter red. Aspiring, which your father takes, is know to cause intestinal and stomach bleeding in many patients. Probably your father should also receive a test for blood hemoglobin to be sure that he has not become seriously anemic from continued blood loss into his bowel and out in his stool (feces).
mayo clinic?, my husband had a heart attack on 11/04/04. he is on several medications,toprol, lipitor,calcitriol,zetia and hyzaar. his heart is fine but he feels sick everyday. is it possible for medications to be toxic for some patients? he has had several test for the vertigo he suffers from and everything is fine according to the doctors. they have told us good luck and sent us on our way. can you help.

kidney disease in dog?, My 16yo poodle/schnouzer mix was diagnosed with kidney disease last yr. Please tell me anything more that I can do as his kidneys keep getting worse. His diet is Hill's Prescription diet for kidneys canned. His meds are: calcitriol 1.5ml and 3 azodyl pills a day for kidneys, Enalapril once daily for blood pressure, and cosequin ds daily for arthritis. I also give him 250ml of normal saline via subcutenous IV now twice a week. His hips and back legs suffer the most from bone loss and arthritis. But the vet said he'sin no pain from the kidney disease. But is there anything more I can do? I bought a bottle at a pet store to force him to drink more since the vet said to flush his kidneys. but theres increased protein in his urine and decreased in his blood. I just want to do all that I can to make him live as healthy as possibl. Should I force exercise should I force water more. Some nights he eats half his food should I force more? What else any other drugs I should give him? Anything pls
My 8 year old Yorkie just had surgery to remove a large kidney stone. He also has an enlarged heart. He will be on the Enalapril for the rest of his life. I feel for you as I too am wondering what can I do to keep him healthy and give him longevity. You are most fortunate to have had him for 16 years. I think you are doing all that you reasonably can at this point. You will 'know' when it's time to end his suffering and let him go, painful as it may be. Just give him all of the love and tenderness you can and God bless you both.
Gastric Bypass & Hypoparathyroidism?, I am scheduled for a gastric bypass on the 22nd of this month. One caveat: I have surgically induced hypoparathyroidism as a result of thyroid CA treatment 6 years ago. My bariatric physician feels this should not be of concern because my calcium levels are well controlled with 1000 mg CA+ daily along with 0.25 mcg of calcitriol daily and 50,000 IU of Vit D 3x week.
I'd appreciate hearing from other medical professionals about this situation, along with others who may have undergone a bypass with co-existing HPTH.
First of All Hypothyroidism and hypoparathyroidism are not the same thing not even remotely related. My hypoparathyroidism was also surgically induced when my throid was taken out. As Long as you show no signs of Tetany and your calcium levels are normal, I would think you will be fine. But be warned if your calcium levels drop you know what can happen so I would talk this over with several physicians before you do this.
My Shih-tzu was diagnosed with early kidney failure and has had diarrhea for 4 days?, My 9 lb. Shih-tzu (almost 14) was diagnosed last Monday with early kidney failure. My vet immediately (Tuesday) gave her fluids under her skin and put her on Calcitriol and Purina NF (which she loved). Then on Wed. night she started with diarrhea and wouldn't eat or drink anything. I took her back to the vet Fri. morning and he could not find a reason for her having diarrhea. He gave her a Centrine injection and VBG 1 ml and put her on 1/2 Centrine 0.2 mg tab every 12 hours. Yesterday the vet switched her food to another for kidney failure (I don't remember the name of it) but she doesn't want it either. The only thing she seems to want is plain turkey or cheese (to get her to take the pill). I also tried the food she used to eat (Iams mini-chunks) but she won't eat it. She is drinking a little water. She has diarrhea every 1 1/2-2 hours and is a very little amount sometimes with a little blood. Any ideas on what's wrong and what I can feed her? Please help! Thank You So Much!
Kidney failure causes lots of stuff to be left in the blood stream that should be cleaned out by the kidneys - and that makes you feel terrible, and everything also tastes bad. It'll be a tough game top find out what appeals to her, if anything.
Ask the vet if theycan give you some probiotic paste to help stabilize the bacteria in her GI tract, and whether she could have some canned pumpkin to help that situation as well.
This is a tough situation, as your dog is very sick, and a lot of things won't work no matter how hard you- and the vet- try. It isn't the vet, it's the disease. I'm really sorry.
