Fetal Risk Summary

Amantadine is used as an anti-Parkinson agent and as an antiviral drug for influenza A treatment and prophylaxis. In a 1975 correspondence, one author thought the drug was a potential human teratogen and the absence of published reports at the time had more to do with its infrequent use in pregnancy than to its teratogenic potency (1).

Amantadine was teratogenic in pregnant rats at 50 mg/kg/day (estimated human equivalent dose [EHED] based on body surface area conversion of 7.1 mg/kg/day) and embryotoxic at 100 mg/kg/day (EHED of 14.2 mg/kg/day) (2). No embryo or fetal harm was observed at 37 mg/kg/day (EHED of 5.3 mg/kg/day). No teratogenic or embryotoxic effects were seen in pregnant rabbits dosed with 32 mg/kg/day (EHED of 9.6 mg/kg/day) (2). The recommended human dose for antiviral indication is 200 mg/day and 100–200 mg/day for Parkinsonism.

No reports describing the placental passage of amantadine in animals or humans have been located. The molecular weight of the drug (approximately 188 for the hydrochloride salt) is low enough, however, that passage to the fetus should be expected.

A cardiovascular defect (single ventricle with pulmonary atresia) has been reported in an infant exposed to amantadine during the 1st trimester (3). The mother was taking 100 mg/day for a parkinson-like movement disorder. The relationship between the drug and the defect is unknown.

In a surveillance study of Michigan Medicaid recipients involving 229,101 completed pregnancies conducted between 1985 and 1992, 51 newborns had been exposed to amantadine during the 1st trimester (F. Rosa, personal communication, FDA, 1993). A total of five (9.8%) major birth defects was observed (two expected). Among six categories of defects for which specific data were available, one cardiovascular defect (0.5 expected) and one limb reduction defect (0 expected) were observed. No cases of oral clefts, spina bifida, polydactyly, or hypospadias were recorded. Although the incidence of defects is high, the number of exposures is too small to draw any conclusions.

Breast Feeding Summary

Amantadine is excreted into breast milk in low concentrations. Although no reports of adverse effects in nursing infants have been located, the previous manufacturer recommended the drug be used with caution in nursing mothers because of the potential for urinary retention, vomiting, and skin rash (4). The current manufacturer recommends that amantadine should not be used in nursing mothers (2).

Can I stop taking amantadine if I feel better?
I was prescribed both amantadine and zithromax for a bad fever/chest cough. I'm done with the zithromax (antibiotic) and know never to stop antibiotics prematurely. Is this true for an antiviral medicine too? If I'm feeling a lot better can I discontinue taking the amantadine, or do I need to take it to completion?

talk to your MD about it. but you should complete your prescription. if you stop prematurely, there is a chance that you create a resistant and it will be harder to treat.

it is an antiviral drug that is used to prevent getting the flu - similar to the recent popular drug called Tamiflu that was publicized for preventing the "bird flu"

another use for amantadine is in Parkinson patients. it can help increase a chemical called dopamine in the brain.

Amantadine can also be used in head injury patients to help with their attention and concentration.

this drug does not have a great deal of side effect and it is usually pretty safe.

no you will not get "tripped". amantadine is used for the flu in elderly patients. this should NOT cause any problems, however, check with your primary care physician.

Can trouble speaking, swallowing, & breathing be symptoms of MS? Or Amantadine? Like the tongue is too big.?
My husband has MS. He is on Betaseron and Amantadine, and is having difficulties with speaking, breathing, and eating; he says it feels like his tongue is huge. Is this more likely to be due to the MS, or the medications?

It could be an allergic reaction to the Betaseron or the Amantadine.

An allergic reaction would include difficulty breathing; closing of the throat; swelling of the lips, tongue, or face; or hives.

Seek medical attention ASAP!

Yeah. Amantadine is medication for the flu. Her kidneys were crushed in the bus accident. Without her kidneys functioning, her body was unable to filter the pills she took just before the crash. Usually dialysis would take care of her problem, but this particular drug is unable to be flushed out of the blood by dialysis. If you were a patient on dialysis, you would never be given this drug because of this complication. Unfortunately, this was just a very horrible coincidence that resulted in Amber's death.

Dog has brownish substance and bitter smell in one ear. Amantadine was first used, and her ears were cleaned, and she still is showing signs of infection. I believe it is probably a yeast infection, and I would like to use some Diflucan to treat her. I don't know doseage for and 85 lb Labrador.

Please contact your vet. He/she is the one qualified to tell you this. Take your dog in to see the vet. He can tell more from seeing her ears. He can also tell you definitely whether or not your doggie has a yeast infection and he can prescribe drops in the correct dosage for her weight and breed.

How to help my boyfriend, who has a very sick dog?
His dog is 14, and was diagnosed with stomach cancer. Since being put on crafate and pepcid, he has had no further bleeding, but is crying with pain over what is presumed to be arthritis. He is stiff. Vet is giving him codeine and amantadine. My BF is trying to do everything to get this dog to hang in there. I made a vague reference to euthanasia, and was harshly scolded. Then he is telling me I am being hostile. All this recent lashing out is baffling, as I have tried only to be supportive (my dog was euthanized last week, on the 31st.). I said to him that he tended to lash out when severely upset or worried about anything, and he hung up on me. I hate when he hangs up on me. He was very angry with me. I only meant to tell him that I am trying to be supportive, and to ask him to understand that I am still grieving my dog, but to please not take it out on me? I think he needs to let the dog go, and that he is in denial. It's hard to watch him...Is all this normal? Hurts!

Both of your reactions are completely normal. The grieving process hits us all differently.

Now is not the time to take what he says or his behavior personally. Also, it's not the time to point out that he's being rude, insensitive, or anything else. It's time for you to be mature, both in yourself (your own personality), and in the relationship. Give him some space, let him come to the sad conclusion on his own, and grieve his beloved pet his own way. Once the dog either recovers or dies and some grieving time passes, then bring up rude, insensitive, and talk about your relationship.

Sorry for your loss and your trouble. I hope everything works out for you, your BF and his dog.

What the heck is Amantadine?

Let me begin explaining this.

It's obviously a prescription pill. I was in a pretty serious car wreck last year in October, and I was prescibed to Amantadine. I was supposed to take one capsule by mouth twice a day.

I have no idea what Amantadine is! I should have done the research a long time ago. I just couldn't find the time. I just now remembered about it.

I found the pill bottle in my bathroom, which it still has some pills in it. I'm not going to flush them yet until I figure out what they heck they did for me!

I guess it might help to know that I have, or had,a TBI. Which stands for Tramautic Brain Injury...

I've been told it's a pill that helps me stay alert to things all the time. I've also heard it's an anti depressant...

I want to honestly know what the heck I was taking for nearly a freaking year!!

What is amantadine?

Amantadine is an antiviral medication. It blocks the actions of viruses in your body.

Amantadine is used to treat and to prevent influenza A (a viral infection).

Amantadine is also used to treat Parkinson's disease and "parkinson-like" symptoms such as stiffness and shaking that may be caused by the use of certain drugs. How amantadine works in these conditions is unknown.
